Restriction Analysis of DNA
In this experiment students learn all about the structure of DNA and how it can be manipulated by using a variety of techniques. The procedure demonstrates the cutting of DNA from the bacteriophage lambda with restriction enzymes. The DNA fragments are then analyzed by agarose gel electrophoresis.
Objectives
- Identify the structure and function of DNA
- Define genetic polymorphism and comprehend how this is exploited for forensic applications
- Explain the activity of restriction endonucleases
- Understand the physics of gel electrophoresis
- Learn techniques for collecting, transferring and loading micro amounts of liquid materials
